Why go after him and other file lockers while not expending resources to take down Manwin?

Because the DOJ has targeted file lockers as not falling under safe harbor when the vast majority of their content is illegal. Whether you want to believe it or not Youtube's vast majority of content is not illegally obtained and transmitted.

The DOJ has not been shown to have applied this same prejudice against Tubes thus far. Also, believe it or not a lot of the content in Manwin's tubes is not illegal. They do behind the scenes deals to license a very large portion of it and own the parent companies of much of it. While there are plenty of smaller companies being screwed by them and not receiving any licensing fees they are actually the minority.

If a file locker tried to work out licensing and charging for downloads the way Manwin has they wouldn't be having problems like Manwin isn't.
